Overview

ATI-502 is an investigational small molecule inhibitor of Janus kinase 1 (JAK1) and Janus kinase 3 (JAK3), developed as a topical formulation for the treatment of various forms of alopecia. Developed by Aclaris Therapeutics, it was specifically designed to modulate the inflammatory signaling pathways—including interferon-gamma and cytotoxic T-cell activity—that contribute to autoimmune-mediated hair follicle destruction. The drug was evaluated as a 0.46% topical solution applied twice daily. While Phase 2 trials (AA-201 and AA-202) demonstrated that the drug could penetrate the skin and influence relevant genetic signatures, the program for alopecia areata was terminated in September 2019 after failing to show a statistically significant improvement over the vehicle (placebo) group, which exhibited high rates of spontaneous resolution.
