Atractylenolide I is a naturally occurring sesquiterpene lactone primarily isolated from the rhizomes of Atractylodes macrocephala, a traditional Chinese medicinal herb. It is one of the principal bioactive constituents of this plant and is known for diverse biological activities, including anti-inflammatory, anti-apoptotic, antioxidant, anti-coagulant, and anti-cancer effects. Mechanistically, atractylenolide I has been shown to inhibit the TLR4/MyD88/NF-κB pathway, downregulate pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6, modulate macrophage polarization, and block the JAK2/STAT3 signaling cascade, resulting in apoptosis induction in cancer cells and suppression of glycolysis. It has been explored experimentally in various disease models, especially for gastrointestinal disorders, sepsis, acute liver failure, and multiple cancers, including colorectal cancer[1][2][3][5][9].
