Autologous anti-CD20 CAR T cells (also known as CART-20) is an investigational cell-based gene therapy developed by the Chinese PLA General Hospital. This adoptive T-cell therapy involves genetically engineering a patient's own T cells to express a chimeric antigen receptor (CAR) that specifically targets the CD20 surface antigen, which is highly expressed on B-cell malignancies. The specific construct used is CAR.20-CD137ζ, which incorporates a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain to enhance T-cell activation and persistence. CART-20 is primarily indicated for the treatment of chemotherapy-resistant or refractory CD20-positive B-cell leukemia and B-cell lymphoma, including non-Hodgkin lymphoma. Clinical trials have demonstrated high objective response rates with a manageable safety profile in patients with advanced B-cell malignancies.
