Overview

Autologous CD19-CD20-NKG2D-nsBicephali CAR-T cells is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed by Beijing Boren Hospital. It is designed for the treatment of relapsed or refractory primary and secondary central nervous system lymphoma (CNSL). The therapy involves engineering a patient's own T cells to express a multi-specific CAR that simultaneously targets CD19, CD20, and ligands of the NKG2D receptor (such as MICA/B and ULBPs). This trivalent targeting strategy is intended to overcome tumor antigen heterogeneity and reduce the risk of immune escape associated with the loss or downregulation of a single target. The 'nsBicephali' designation refers to the specific structural architecture of the CAR construct. These engineered cells are designed to migrate into the cerebrospinal fluid and brain parenchyma to directly eliminate malignant B cells within the central nervous system.
