Autologous CD20-specific T cells is a chimeric antigen receptor (CAR) T-cell therapy developed by the Fred Hutchinson Cancer Center for the treatment of various B-cell malignancies. The therapy involves the genetic modification of a patient's own T cells to express a CAR targeting the CD20 antigen, a protein widely expressed on the surface of B-cell lymphomas. In clinical trials, these modified cells are typically administered following lymphodepletion with cyclophosphamide and are sometimes accompanied by aldesleukin (IL-2) to support T-cell expansion and persistence. This therapy has shown activity in relapsed or refractory indolent B-cell non-Hodgkin lymphomas, including follicular lymphoma and mantle cell lymphoma. An optimized third-generation version of this construct, incorporating both CD28 and 4-1BB costimulatory domains, is being further developed as MB-106 through a licensing agreement with Mustang Bio.
