Overview

Azacitidine + golidocitinib + CHOP is an investigational combination regimen being evaluated for the frontline treatment of newly diagnosed peripheral T-cell lymphoma (PTCL). This regimen utilizes a genotype-guided strategy, combining the hypomethylating agent azacitidine (a DNA methyltransferase inhibitor), the selective JAK1 inhibitor golidocitinib, and the standard CHOP chemotherapy backbone (cyclophosphamide, doxorubicin, vincristine, and prednisone). Azacitidine is intended to act as an epigenetic primer, potentially sensitizing lymphoma cells to chemotherapy by inhibiting DNA methylation. Golidocitinib specifically targets the JAK1/STAT signaling pathway, which is frequently dysregulated in T-cell malignancies. The combination is currently being studied in a Phase I/II clinical trial led by Ruijin Hospital to determine the recommended Phase II dose and evaluate efficacy compared to standard CHOP.
