Overview

B7-H3 siRNA (also known as CD276 siRNA) is a research-stage RNA interference (RNAi) agent designed to silence the expression of B7-H3, a transmembrane protein and member of the B7 family of immune regulators. B7-H3 is frequently overexpressed in various malignancies, including breast cancer and melanoma, where it contributes to tumor cell proliferation, invasion, and resistance to chemotherapy. Research presented at AACR 2016 demonstrated that knockdown of B7-H3 using siRNA reduces the glycolytic capacity of tumor cells and increases their sensitivity to both traditional anti-cancer agents and targeted inhibitors of the PI3K/AKT/mTOR and MAPK pathways. This suggests that B7-H3 plays a critical role in metabolic reprogramming and therapeutic resistance, making its suppression a potential strategy for enhancing the efficacy of existing cancer treatments.
