B7-H3-targeted CAR T-cell therapy is an investigational cell-based immunotherapy designed to treat various solid and hematologic malignancies. It involves genetically engineering T cells (either autologous or allogeneic) to express a chimeric antigen receptor (CAR) that specifically recognizes B7-H3 (CD276), an immune checkpoint molecule frequently overexpressed in tumors but with limited expression in healthy tissues. Upon binding to B7-H3 on the surface of cancer cells, these engineered T cells are activated to exert potent cytotoxic effects. This therapeutic approach is being explored across a wide range of indications, including glioblastoma, pediatric solid tumors, and acute myeloid leukemia, with several distinct constructs currently in early-phase clinical and preclinical development.
