B7-H3-targeted CAR-T cells co-expressing 4-1BBL-OX40L fusion protein is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to treat solid tumors. Developed by Fuzhou Tcelltech Biological Science and Technology, the therapy targets B7-H3 (CD276), a checkpoint molecule frequently upregulated in malignancies such as glioblastoma and colon cancer. To overcome the immunosuppressive microenvironment and limited persistence typical of solid tumor treatments, these CAR-T cells are engineered to co-express a membrane-bound fusion protein of 4-1BB ligand (4-1BBL) and OX40 ligand (OX40L). This dual costimulation aims to enhance T-cell activation, proliferation, and survival while improving tumor infiltration and reducing exhaustion. Preclinical studies have demonstrated that this construct exhibits superior anti-tumor activity and persistence compared to standard B7-H3 CAR-T cells.
