Overview

B7-H3 targeting CAR T cells are an investigational cell therapy modality designed to treat solid tumors by targeting the CD276 (B7-H3) antigen. B7-H3 is a transmembrane protein in the B7 family that is highly expressed in various malignancies, including glioblastoma, neuroblastoma, and several carcinomas, while having limited expression in normal tissues. These CAR T cells are engineered to recognize B7-H3, triggering T-cell activation and direct cytolysis of tumor cells. Advanced iterations of this platform, such as those developed by researchers at Memorial Sloan Kettering Cancer Center and Weill Cornell Medicine, incorporate armoring strategies—such as the secretion of PD-1 blocking scFvs—to counteract immunosuppressive signals in the tumor microenvironment and enhance persistence and efficacy in challenging indications like glioblastoma.
