Overview

B7H3-targeting CAR T cells + CTSR-targeting CAR T cells is a dual-targeting chimeric antigen receptor (CAR) T-cell therapy developed by the University of Southern California and Children's Hospital Los Angeles. This therapeutic approach involves the simultaneous transduction of T cells with two distinct lentiviral vectors: one encoding a CAR specific for B7-H3 (CD276) and another encoding a Constitutive TLR Signaling Receptor (CTSR). B7-H3 is a checkpoint molecule frequently overexpressed in various solid tumors, including pediatric cancers such as neuroblastoma and medulloblastoma. The CTSR component is designed to provide constitutive toll-like receptor signaling, which serves to enhance T-cell expansion, persistence, and effector function within the typically immunosuppressive tumor microenvironment. The manufacturing process has been optimized using G-Rex static culture devices to ensure linear scalability for clinical production.
