baricitinib + tofacitinib

Baricitinib and tofacitinib are both oral Janus kinase (JAK) inhibitors approved for the treatment of rheumatoid arthritis. They work by inhibiting JAK enzymes involved in pro-inflammatory signaling pathways. Baricitinib (Olumiant) is a selective JAK1/2 inhibitor with moderate activity against TYK2 and limited activity against JAK3. Tofacitinib (Xeljanz) is a selective JAK1/3 inhibitor with limited inhibitory effect on JAK2 and TYK2. Both drugs have demonstrated rapid improvements in disease activity, function, and patient-reported outcomes in rheumatoid arthritis patients. They are typically used in patients who have had an inadequate response to one or more conventional synthetic DMARDs.
