BaseCAR-01 is an autologous, bispecific chimeric antigen receptor (CAR) T-cell therapy developed by the University Hospital Basel for the treatment of B-cell malignancies. The therapy is engineered to simultaneously target two distinct B-cell surface antigens, CD19 and CD20, a strategy designed to reduce the risk of relapse caused by antigen escape or downregulation. The manufacturing process is conducted locally at the hospital's GMP facility, where a patient's T cells are collected via lymphapheresis, genetically modified to express the bispecific CAR, and expanded. Following lymphodepleting chemotherapy (typically cyclophosphamide and fludarabine or bendamustine), the personalized cells are re-infused into the patient. BaseCAR-01 is currently being evaluated in a Phase I clinical trial for adult patients with relapsed or refractory B-cell lymphoma or leukemia who have exhausted standard treatment options or lack access to commercial CAR-T products.
