BB-031 is a first-in-class RNA aptamer that targets the A1 domain of von Willebrand Factor (vWF), an important structural component of blood clots. It acts as a thrombolytic agent by inhibiting vWF, thereby preventing platelet adhesion and aggregation, which enables rapid recanalization of blocked blood vessels in acute thrombotic conditions such as ischemic stroke. The drug is designed for rapid onset and short duration of action, with its effects quickly neutralizable by a complementary reversal agent (BB-025) in case of bleeding. Developed by Basking Biosciences, BB-031 has demonstrated safety and tolerability in Phase 1 trials with healthy volunteers and is currently being evaluated in Phase 2 clinical trials for acute ischemic stroke. Preclinical studies have also shown potential efficacy in pulmonary embolism[1][2][3][4][5][7].
