BEBT-908 + rituximab + gemcitabine + oxaliplatin is an investigational combination regimen for the treatment of relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL). BEBT-908 (Ifupinostat) is a dual inhibitor targeting phosphoinositide 3-kinase (PI3K) and histone deacetylase (HDAC), showing promising anti-tumor efficacy in hematological malignancies through c-Myc downregulation and ferroptosis induction[1][4][5]. Rituximab is a monoclonal antibody targeting CD20 on B cells, causing cell lysis primarily via complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. Gemcitabine is a nucleoside analog acting as an antimetabolite to inhibit DNA synthesis. Oxaliplatin is a platinum-based chemotherapeutic causing DNA cross-linking and apoptosis. When combined (R-GemOx + BEBT-908), these agents employ multi-modal antineoplastic activity: epigenetic modulation, kinase inhibition, immunotherapy, and cytotoxic chemotherapy[4][5]. The combination regimen aims to overcome resistance and improve outcomes in patients who failed previous therapies.
