BHV-1 is a novel oncolytic viral immunotherapy based on bovine herpesvirus type 1 (BoHV-1), currently in preclinical development by 3io Therapeutics in partnership with McMaster University. The therapy is designed to treat KRAS-mutated cancers, including lung, colorectal, and pancreatic cancers. Unlike traditional oncolytic viruses that rely on direct viral replication and tumor lysis, BHV-1 functions as an immuno-stimulating platform whose efficacy is primarily driven by the stimulation of anti-tumor immunity. The virus is non-pathogenic in humans and has demonstrated the potential for systemic intravenous (IV) delivery, maintaining its efficacy even in the presence of neutralizing antibodies. Preclinical studies have shown that BHV-1 can significantly boost the efficacy of immune checkpoint inhibitors. The platform allows for monotherapy with minimal genetic engineering, although cassette payload insertion is also possible for enhanced therapeutic effects.
