Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—bleomycin, etoposide, and methotrexate. Each drug has a distinct mechanism of action: - Bleomycin is an antitumor antibiotic that induces DNA strand breaks through free radical formation. - Etoposide is a topoisomerase II inhibitor that prevents DNA re-ligation during replication, leading to cell death. - Methotrexate is an antimetabolite and antifolate agent that inhibits dihydrofolate reductase, blocking DNA synthesis. The combination has been used as salvage therapy for gestational trophoblastic disease resistant to other multiagent regimens and in the treatment of certain germ cell tumors. Clinical studies have shown high rates of complete remission in patients with drug-resistant choriocarcinoma and gestational trophoblastic neoplasia[1][2][5]. The regimen may be referred to as MBE (methotrexate, bleomycin, etoposide) in some literature.
