BLU-924 (also known as SAR449336) is an orally bioavailable, potent, and selective pan-KRAS inhibitor developed by Blueprint Medicines in collaboration with Sanofi. It is designed to target multiple KRAS mutations, including G12D, G12V, and G13D, as well as the wild-type KRAS protein. By inhibiting KRAS, BLU-924 disrupts the MAPK signaling pathway, which is frequently dysregulated in various solid tumors. It is currently being evaluated in Phase 1/2 clinical trials for the treatment of advanced pancreatic ductal adenocarcinoma, non-small cell lung cancer, and colorectal cancer harboring KRAS mutations, both as a monotherapy and in combination with other agents.
