This is a combination therapy consisting of BMS-275291 (rebimastat), carboplatin, and paclitaxel. BMS-275291 is an orally administered, broad-spectrum matrix metalloproteinase inhibitor (MMPI) developed to inhibit tumor growth and metastasis by blocking the activity of matrix metalloproteinases involved in extracellular matrix degradation. Unlike earlier MMPIs, it was designed to avoid dose-limiting joint toxicity. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis in cancer cells. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division by preventing microtubule depolymerization. This combination was investigated primarily for advanced non-small-cell lung cancer (NSCLC). Clinical trials found that adding BMS-275291 to standard chemotherapy with carboplatin and paclitaxel increased toxicity without improving survival outcomes in NSCLC patients[1][3][4].
