BMS-986416 is an investigational allogeneic (off-the-shelf) chimeric antigen receptor (CAR) T-cell therapy designed to target CD19-positive B cells. Developed by Bristol Myers Squibb, it is being evaluated for the treatment of severe B-cell-mediated autoimmune diseases, such as systemic lupus erythematosus (SLE). Unlike autologous CAR-T therapies that require harvesting a patient's own cells, BMS-986416 utilizes T-cells from healthy donors, which are engineered to express a CAR that recognizes CD19. This approach aims to achieve deep and rapid depletion of pathogenic B cells and provide an "immune system reset." The therapy is currently in Phase 1 clinical development to assess its safety, tolerability, and initial efficacy in patients with refractory autoimmune conditions.
