Overview

**BMS-986458 + mosunetuzumab** is an investigational combination therapy comprised of two components: BMS-986458, an orally available, highly selective cereblon (CRBN) E3 ubiquitin ligase-dependent heterobifunctional ligand-directed degrader of B-cell lymphoma 6 (BCL6), and mosunetuzumab, a bispecific monoclonal antibody that engages CD3 on T cells and CD20 on B cells to redirect T cells for targeted B-cell lymphoma cell lysis. BMS-986458 acts by degrading BCL6 to induce anti-tumor activity and upregulate CD20 expression, while mosunetuzumab functions as a T-cell engager facilitating immune-mediated clearance of malignant B cells. This combination is under investigation for the treatment of relapsed or refractory non-Hodgkin lymphoma in clinical trials, aiming to offer a chemotherapy-free regimen for B-cell malignancies[2][4].
