BPI-585771 is an orally bioavailable small molecule inhibitor of the KRAS protein, currently under development by Betta Pharmaceuticals. KRAS is a member of the RAS family of GTPases and acts as a critical molecular switch in signaling pathways that regulate cell growth and survival, such as the MAPK/ERK and PI3K/AKT pathways. Mutations in the KRAS gene are among the most frequent oncogenic drivers in human cancers, particularly in non-small cell lung cancer (NSCLC), colorectal cancer, and pancreatic adenocarcinoma. BPI-585771 is designed to bind to and inhibit the activity of the KRAS protein, thereby disrupting downstream oncogenic signaling and suppressing tumor cell proliferation. As of early 2024, the candidate is in the IND-enabling stage of development for the treatment of advanced solid tumors.
