Overview

This is a five-drug combination regimen consisting of brentuximab vedotin, doxorubicin, vinblastine, dacarbazine, and rituximab. Brentuximab vedotin is an antibody-drug conjugate targeting CD30 on Hodgkin lymphoma cells; it delivers the cytotoxic agent monomethyl auristatin E (MMAE) to malignant cells. Doxorubicin is an anthracycline chemotherapeutic that intercalates DNA and inhibits topoisomerase II. Vinblastine is a vinca alkaloid that disrupts microtubule formation during mitosis. Dacarbazine acts as an alkylating agent causing DNA damage. Rituximab is a monoclonal antibody targeting CD20 on B lymphocytes, leading to cell lysis via immune-mediated mechanisms. This combination has been studied in children, adolescents, and young adults with newly diagnosed classic Hodgkin lymphoma (cHL), showing high complete response rates and favorable survival outcomes[2].
