Overview

Brentuximab vedotin + rituximab is a combination of two monoclonal antibody-based therapies used primarily in the treatment of relapsed or refractory large B-cell lymphoma (LBCL), including diffuse large B-cell lymphoma (DLBCL). Brentuximab vedotin is an antibody-drug conjugate targeting CD30, delivering the cytotoxic agent monomethyl auristatin E (MMAE) to tumor cells. Rituximab is a CD20-directed monoclonal antibody that targets CD20 on B lymphocytes, leading to their elimination by immune-mediated mechanisms. The combination of these agents exerts both direct cytotoxic and immunomodulatory effects on malignant B cells. The combination is typically studied and administered with other agents, most notably lenalidomide, but data also exists for use as a dual combination in certain contexts. Both drugs are developed and manufactured by major pharmaceutical companies, and have established roles in immunotherapy for hematologic malignancies[5][9].
