Overview
This is a **three-drug combination regimen** composed of brentuximab vedotin (an antibody-drug conjugate targeting CD30), rituximab (a monoclonal antibody targeting CD20), and bendamustine (a bifunctional alkylating agent). The combination is being investigated primarily for the treatment of relapsed or refractory CD30-positive diffuse large B-cell lymphoma (DLBCL) as salvage therapy, particularly for patients ineligible for autologous stem cell transplant. Brentuximab vedotin delivers the microtubule-disrupting agent MMAE via anti-CD30 targeting, causing apoptosis of CD30-expressing tumor cells. Rituximab targets the CD20 antigen on B-cells, mediating B-cell depletion via multiple cytotoxic mechanisms. Bendamustine induces DNA crosslinking and damage in lymphocyte populations, inhibiting tumor proliferation.
