Brentuximab vedotin is an **antibody-drug conjugate (ADC)** targeting **CD30**, delivering the microtubule disruptor monomethyl auristatin E (MMAE) to induce cell death in CD30-expressing tumor cells. Rituximab is a **monoclonal antibody** targeting **CD20** on B-cells, promoting antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and apoptosis. Nivolumab is a **PD-1 inhibitor** that blocks the PD-1/PD-L1 interaction, enhancing T-cell mediated anti-tumor immunity. This triplet leverages targeted delivery, B-cell depletion, and immune checkpoint blockade, showing promise in frontline and relapsed settings for B-cell malignancies with CD30 and CD20 expression.[1]
