Overview

Brodalumab is a fully human monoclonal IgG2 antibody used for the treatment of moderate to severe plaque psoriasis in adults who are candidates for systemic therapy or phototherapy and have not responded adequately to other treatments. It works by selectively binding to interleukin 17 receptor A (IL-17RA), thereby inhibiting its interaction with several pro-inflammatory cytokines from the IL-17 family, including IL-17A, IL-17F, IL-17C, the IL‑17A/F heterodimer, and IL‑25. By blocking these interactions, brodalumab inhibits downstream inflammatory responses implicated in the pathogenesis of psoriasis[2][4][6][8]. Brodalumab is administered as a subcutaneous injection and is available under the brand name Siliq. Due to observed risks of suicidal ideation and behavior, it is only available through a restricted program under FDA’s Risk Evaluation and Mitigation Strategies (REMS)[7].
