BVB808 is a small molecule Janus kinase (JAK) inhibitor developed by Novartis. It has been primarily investigated in preclinical research for the treatment of multiple myeloma, often in combination with other targeted agents like the FGFR inhibitor BGJ398 (infigratinib). The compound targets the JAK-STAT pathway, which is a critical mediator of cytokine-induced survival signals (such as IL-6) in the bone marrow microenvironment. Preclinical studies have demonstrated that BVB808 can synergize with FGFR inhibitors to overcome resistance and enhance anti-tumor activity in specific genetic subsets of multiple myeloma, such as those with the t(4:14) translocation and FGFR3 mutations.
