C-CAR168 is a novel autologous bispecific chimeric antigen receptor T (CAR-T) cell therapy developed for the treatment of autoimmune diseases, particularly resistant and refractory Lupus Nephritis (LN) and Systemic Lupus Erythematosus (SLE). It is engineered to target both CD20 and B-cell maturation antigen (BCMA), which are markers associated with pathogenic B cells and long-lived plasma cells implicated in autoimmune responses. By targeting these antigens, C-CAR168 aims to deplete disease-causing B cells, plasmablasts, and plasma cells—including age-associated B cell subsets—thereby reducing autoantibody production. Preclinical studies have demonstrated potent cytotoxicity against CD20- or BCMA-positive cells with minimal off-target toxicity. The drug is currently in early-phase clinical trials for LN/SLE and may be expanded to other autoimmune indications such as Neuromyelitis Optica Spectrum Disorder (NMOSD), Immune-Mediated Necrotizing Myopathy (IMNM), and Systemic Sclerosis (SSc)[1][2][3][4][5][7].
