Overview

CA-STAT5 refers to a constitutively active mutant form of the Signal Transducer and Activator of Transcription 5 (STAT5) protein. It is primarily utilized in research and preclinical development as a genetic modification for adoptive cell therapies (ACT), such as TCR-engineered T cells. By providing continuous STAT5 signaling independent of external cytokine stimulation (e.g., IL-2 or IL-15), CA-STAT5 enhances T-cell expansion, persistence, and polyfunctionality. Research indicates that CA-STAT5-engineered T cells demonstrate superior antitumor activity in models like B16F10 melanoma by reducing PD-1 expression and attenuating the PD-1/PD-L1 inhibitory pathway, thereby decreasing cellular exhaustion. Additionally, it is used as a laboratory tool to investigate the role of STAT5 signaling in dendritic cell differentiation and the pathogenesis of myeloid neoplasms.
