Overview

A combination therapy comprising camrelizumab, a humanized monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, and ivarmacitinib (also known as HMPL-590), a selective small-molecule inhibitor of Janus kinase 1 (JAK1). Camrelizumab acts as an immune checkpoint inhibitor, restoring the ability of T cells to recognize and attack tumor cells by blocking the PD-1/PD-L1 interaction. Ivarmacitinib modulates the tumor microenvironment and cytokine signaling pathways that contribute to tumor growth and immune evasion. This combination is currently being evaluated in clinical trials, specifically as a first-line treatment for pediatric patients with classical Hodgkin lymphoma (cHL). The rationale for the combination is that PD-1 blockade relieves immune suppression, while JAK1 inhibition further modulates the inflammatory environment characteristic of cHL.
