CAR-T cells targeting B7-H3 and PTGFRN are an experimental chimeric antigen receptor T-cell therapy being developed for the treatment of glioblastoma. B7-H3 (CD276) and PTGFRN (Prostaglandin F2 receptor inhibitor) were identified as tumor-specific surface antigens highly expressed in glioblastoma surgical specimens but not in normal brain tissue. In preclinical studies conducted at Osaka University, these CAR-T cells demonstrated significant anti-tumor activity, including increased production of IL-2 and IFN-γ upon co-culture with GBM cells and reduced tumor growth in orthotopic xenograft models. The therapy aims to overcome the challenges of glioblastoma treatment, such as the blood-brain barrier and the immunosuppressive tumor microenvironment.
