CAR-T cells targeting CD20 are an autologous or allogeneic cellular immunotherapy consisting of T cells genetically engineered to express a chimeric antigen receptor (CAR) directed against the B-lymphocyte antigen CD20. CD20 is a transmembrane protein expressed on the surface of mature B cells and most B-cell malignancies, making it a viable target for treating B-cell non-Hodgkin lymphomas (NHL) and certain leukemias. Upon binding to CD20-expressing tumor cells, the CAR-T cells undergo activation and expansion, leading to targeted cytotoxicity against the malignant cells. This modality is often explored as a sequential or combination therapy alongside CD19- or CD22-targeted CAR-T cells to overcome antigen escape and improve durable remission rates in refractory or relapsed B-cell malignancies.
