Overview

CAR19.20.22 is a tri-specific chimeric antigen receptor (CAR) T-cell therapy developed by the University of Maryland, Baltimore, in collaboration with Miltenyi Biotec. This investigational cell therapy is engineered to target three distinct antigens—CD19, CD20, and CD22—on the surface of B-cell malignancies. By employing a multi-antigen targeting approach, the therapy aims to enhance the immune system's ability to recognize and destroy cancer cells while significantly reducing the risk of antigen-escape relapse, a common limitation of single-target CAR T therapies. The construct also incorporates ICOS (inducible T-cell co-stimulator) and OX40 co-stimulatory domains to improve T-cell activation, persistence, and survival. It is currently being evaluated in a Phase 1a clinical trial for patients with relapsed or refractory B-cell lymphomas and chronic lymphocytic leukemia (CLL).
