Overview

CAR20.19.22 is a trispecific chimeric antigen receptor T cell (CAR-T) therapy designed to target three B-cell antigens—CD19, CD20, and CD22—simultaneously on malignant B cells[2][3]. The therapy uses autologous T cells that are genetically modified with a lentiviral vector encoding a tandem CAR construct capable of recognizing all three targets[1][2]. This multi-targeted approach aims to reduce the risk of antigen escape and improve efficacy in treating relapsed or refractory B-cell malignancies such as non-Hodgkin lymphoma (NHL) and chronic lymphocytic leukemia (CLL)[2][5]. The drug was developed by Miltenyi Biomedicine GmbH and has reached Phase 1 clinical trials but is currently discontinued for further development in these indications[2][5].
