Overview

CART20-NIS DKO is an experimental, human-derived chimeric antigen receptor T (CAR-T) cell therapy designed to target canine CD20 (cCD20). Developed by the Mayo Clinic and LEAH Laboratories, this therapy is engineered with a double knockout (DKO) of the TRAC and B2M genes to reduce xenogeneic rejection and potential graft-versus-host disease. Additionally, it incorporates the sodium iodide symporter (NIS), which serves as a reporter gene allowing for real-time, non-invasive monitoring of CART cell expansion and trafficking via PET imaging (using tracers like 18F-TFB). The construct utilizes a 4-1BB costimulatory domain. It has been evaluated in companion dogs with spontaneous diffuse large B-cell lymphoma (DLBCL) as a model for human disease and as a veterinary therapeutic.
