CC-122 + CC-223 + rituximab is a triple combination regimen investigated in phase Ib trials for relapsed/refractory diffuse large B-cell lymphoma (DLBCL). - **CC-122 (avadomide)** is a cereblon E3 ligase modulator (CELMoD). It binds cereblon in the cullin 4 E3 ubiquitin ligase complex, promoting ubiquitination and degradation of hematopoietic transcription factors Aiolos and Ikaros. This leads to increased apoptosis of malignant B cells and activation of T cells, resulting in preliminary antitumor activity[1][3]. - **CC-223** is a selective, ATP-competitive mammalian target of rapamycin (mTOR) kinase inhibitor, inhibiting both the mTORC1 and mTORC2 complexes, and is intended to block growth and proliferation signals in malignant cells[1]. - **Rituximab** is an anti-CD20 monoclonal antibody that mediates B-cell lysis via antibody-dependent cellular cytotoxicity, complement activation, and direct apoptosis[1]. This combination was assessed alongside other regimens in the phase Ib CC-122-DLBCL-001 trial (NCT02031419), but the triplet with CC-122 + CC-223 + rituximab was associated with enhanced toxicity and insufficient improvement in efficacy versus doublet regimens.
