Overview

CC-96673 is a humanized IgG1 bispecific monoclonal antibody that co-targets CD47 and CD20. It is designed to bind CD20 with high affinity and CD47 with optimally lowered affinity, allowing selective targeting of CD20-expressing cells such as malignant B cells. By blocking the interaction between CD47 and signal regulatory protein alpha (SIRPα), it disrupts the "don't eat me" signal that protects tumor cells from macrophage-mediated phagocytosis. This dual targeting enhances anti-tumor activity compared to anti-CD20 antibodies alone while reducing off-tumor toxicity associated with broad CD47 inhibition. Preclinical studies have shown favorable pharmacokinetics and toxicity profiles, supporting its development for the treatment of relapsed or refractory non-Hodgkin's lymphoma[1][3][2]. The drug is being developed and manufactured by Bristol Myers Squibb.
