Overview

CD19 CAR T-cell therapy is a form of adoptive cell immunotherapy where a patient's (autologous) or donor's (allogeneic) T cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD19 protein. CD19 is a cell surface marker predominantly expressed on B cells and their precursors, making it an ideal target for B-cell malignancies. Upon infusion, these engineered T cells recognize and eliminate CD19-positive cells through direct cytotoxicity. Several products have received FDA approval for hematologic malignancies, including tisagenlecleucel, axicabtagene ciloleucel, brexucabtagene autoleucel, and lisocabtagene maraleucel. Beyond oncology, CD19 CAR T cells are being investigated for the treatment of severe autoimmune diseases like systemic lupus erythematosus (SLE) and systemic sclerosis (SSc) by inducing deep B-cell depletion.
