Cellogen Therapeutics is developing a bispecific CAR T cell therapy that simultaneously targets CD19 and CD20, two surface markers expressed on B-cell malignancies. This investigational cell therapy is designed to address two major challenges with current CD19-targeted CAR T therapies: antigen-mediated relapse (where patients experience CD19 antigen loss after treatment) and lack of CAR T cell persistence. The therapy utilizes a bispecific CAR construct that incorporates dual costimulatory domains (combinations of CD28, 41BB, ICOS, and OX40) to enhance anti-tumor activity and improve CAR T cell persistence. It is currently in preclinical development for the treatment of Acute Lymphoblastic Leukemia (ALL) and Diffuse Large B-Cell Lymphoma (DLBCL).
