CD20 bispecific antibody-armed activated T cells (CD20Bi-AATC) represent an adoptive cellular immunotherapy platform where autologous T cells are activated ex vivo and then coated with a bispecific antibody (BsAb) before being infused into the patient. The bispecific antibody used is typically a chemical heteroconjugate of anti-CD3 (such as OKT3) and anti-CD20 (such as rituximab). This "arming" process allows the T cells to bypass the MHC-restriction of the T-cell receptor, directly bridging the cytotoxic T cell to CD20-expressing target cells, such as malignant B cells or clonogenic myeloma precursor cells. Unlike CAR-T cells, these cells are not genetically modified but rely on the exogenous bispecific antibody to provide specificity and trigger activation upon contact with the target antigen. The therapy has been primarily investigated in the context of B-cell malignancies and as a consolidative treatment in multiple myeloma patients undergoing autologous stem cell transplantation.
