CD20 CAR (Chimeric Antigen Receptor) therapy represents a class of adoptive cell therapies, primarily CAR-T cells, engineered to express a synthetic receptor targeting the B-lymphocyte antigen CD20. CD20 is a transmembrane protein expressed on the surface of mature B cells and most B-cell malignancies, making it a validated target for immunotherapy. While CD19-directed CAR-T therapies are more widely established, CD20 CAR-T cells are being developed to address CD19-negative relapses (antigen escape) or as part of multi-antigen (e.g., CD19/CD20 bispecific) CAR constructs. The mechanism involves the ex vivo genetic modification of T cells to express a single-chain variable fragment (scFv) specific for CD20, linked to intracellular signaling domains (typically CD3-zeta and a co-stimulatory domain like 4-1BB or CD28), which triggers T-cell activation and tumor cell lysis upon target recognition.
