CD20 x CD3 bispecific antibody is a novel immunotherapeutic agent developed by the University of Birmingham for the treatment of relapsed and refractory mature B-cell non-Hodgkin lymphoma. This bispecific monoclonal antibody is designed to engage the patient's own immune system by simultaneously binding to the CD20 antigen, which is highly expressed on the surface of malignant B-cells, and the CD3 receptor complex on T-cells. By bringing cytotoxic T-cells into direct proximity with cancerous B-cells, the antibody facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent release of perforins and granzymes. This process results in the targeted destruction of the malignant B-cells through T-cell mediated cytotoxicity. This T-cell redirecting strategy aims to provide a potent treatment option for patients who have failed standard therapies, such as rituximab-based regimens.
