Anti-CD24 antibodies are a class of investigational biologics designed to target CD24 (Cluster of Differentiation 24), a GPI-anchored glycoprotein that is highly expressed in various solid tumors and hematological malignancies. CD24 serves as a potent "do-not-eat-me" signal by interacting with Siglec-10 on tumor-associated macrophages, which inhibits phagocytosis and allows cancer cells to evade the innate immune system. By blocking the CD24/Siglec-10 axis, these antibodies promote macrophage-mediated phagocytosis and enhance anti-tumor immunity. Research is currently exploring various formats, including naked monoclonal antibodies, antibody-drug conjugates (ADCs), and bispecific T-cell engagers, with specific interest in indications such as ovarian cancer, triple-negative breast cancer, and glioblastoma.
