Overview

CD47 antagonist peptide refers to a class of peptide-based or peptidomimetic therapeutics designed to disrupt the interaction between CD47, a transmembrane protein overexpressed on various cancer cells, and Signal Regulatory Protein Alpha (SIRPα) located on macrophages. CD47 serves as a "don't eat me" signal that allows tumor cells to evade immune surveillance by inhibiting phagocytosis. By antagonizing this interaction, these peptides promote the macrophage-mediated engulfment of tumor cells. Developed by companies such as Aurigene Discovery Technologies, these antagonists have demonstrated significant preclinical efficacy, including the inhibition of primary tumor growth and lung metastasis in syngeneic mouse models like B16F10 melanoma. Unlike many monoclonal antibodies targeting the same pathway, some of these peptide-based leads exhibit favorable ADME properties and moderate oral bioavailability.
