CD8-targeted lipid nanoparticle delivering CD20 CAR mRNA is an investigational in vivo CAR-T cell therapy. It utilizes targeted lipid nanoparticles (tLNPs) decorated with anti-CD8 binders to selectively deliver mRNA encoding a CD20-directed chimeric antigen receptor (CAR) to endogenous CD8+ T cells. Once the mRNA is translated, the recipient T cells express the CAR and can target and deplete CD20-expressing B cells. This approach aims to provide the efficacy of CAR-T therapy without the need for complex and costly ex vivo cell manufacturing. It is being developed for B-cell malignancies and B-cell-mediated autoimmune diseases such as lupus and myositis.
