Overview

CDNP-R848 is an experimental nanoparticle-based immunotherapy designed to treat glioblastoma by modulating the tumor microenvironment. It consists of a beta-cyclodextrin nanoparticle (CDNP) carrier encapsulating resiquimod (R848), a potent dual agonist of Toll-like receptors 7 and 8 (TLR7/8). The formulation is specifically engineered to target tumor-associated macrophages (TAMs) and other myeloid cells, inducing a pro-inflammatory shift (M1-like phenotype) and enhancing anti-tumor immunity. Preclinical studies in glioma models have shown that CDNP-R848 can induce significant tumor regression and improve survival, particularly when combined with other therapies like VEGF inhibition or anti-CTLA-4 antibodies. The nanoparticle delivery system helps overcome the blood-brain barrier and reduces systemic off-target effects associated with free TLR agonists.
