CGT1815 is a selective, small molecule pan-KRAS(ON) inhibitor being developed by Cogent Biosciences for the treatment of solid tumors harboring KRAS mutations, including colorectal cancer, non-small cell lung cancer, and pancreatic cancer. It is designed as a prodrug of CGT1263, a potent KRAS(ON/OFF) inhibitor that binds to the Switch II pocket of the KRAS protein. CGT1815 demonstrates high selectivity for mutant KRAS over HRAS and NRAS and has shown sub-nanomolar potency across various KRAS mutant cell lines in preclinical studies. The compound is currently undergoing IND-enabling studies with an Investigational New Drug application planned for 2026.
