Overview

CGT6737 is a preclinical, orally administered small molecule developed by Cogent Biosciences for the treatment of cancer. It is described as a pan-KRAS(ON) inhibitor with selectivity over HRAS and NRAS, demonstrating picomolar activity across prevalent KRAS mutations. In preclinical mouse xenograft models, CGT6737 achieved robust pharmacokinetic/pharmacodynamic (PK/PD) profiles and 90% pathway inhibition, resulting in significant tumor growth inhibition. The drug is being optimized to target cancers driven by KRAS mutations, which are among the most common oncogenic drivers in malignancies such as colorectal cancer, non-small cell lung cancer, and pancreatic cancer.
