Chidamide + rituximab + polatuzumab vedotin (CRP regimen) is an investigational combination therapy being evaluated for the treatment of diffuse large B-cell lymphoma (DLBCL), specifically in elderly patients with previously untreated double-positive disease. The regimen combines chidamide, an oral subtype-selective histone deacetylase (HDAC) inhibitor; rituximab, a chimeric monoclonal antibody targeting CD20; and polatuzumab vedotin, an antibody-drug conjugate (ADC) targeting CD79b. This multi-modal approach aims to enhance anti-tumor activity through epigenetic modulation, antibody-dependent cellular cytotoxicity, and targeted delivery of the microtubule-disrupting agent monomethyl auristatin E (MMAE). The regimen is currently being studied in a Phase II clinical trial sponsored by Tianjin Medical University Cancer Institute and Hospital.
